MOTS-c

A profile on the Mitochondrial-Derived Peptide known as an "exercise mimetic" for its ability to regulate metabolism and insulin sensitivity.

MOTS-c

MOTS-c (Mitochondrial ORF of the Twelve S-c) is a peptide encoded in the mitochondria, not the nucleus. It is one of the few "Mitochondrial-Derived Peptides" (MDPs).

It acts as a powerful regulator of metabolism. It has been shown to prevent diet-induced obesity and insulin resistance in mice, earning it the title of an "exercise mimetic."

Active CompoundMOTS-c
Half-Life~30-60 Minutes
MechanismFolate Cycle / AMPK Activation
AdministrationSubcutaneous (SubQ)

Mechanism of Action

MOTS-c translocates to the nucleus where it responds to metabolic stress. It inhibits the folate cycle and activates AMPK (Adenosine Monophosphate-Activated Protein Kinase).

Activation of AMPK is typically what happens during exercise. It tells the body to burn glucose and fatty acids for energy.

Pharmacokinetics & Dosing

Dosing Protocol

MOTS-c dosing is unique and often follows a specific protocol to avoid injection pain (it can sting).

  • Dosage: 5mg - 10mg per injection. (This is a large volume).
  • Frequency: Once Weekly.
  • Protocol: Many users run a 4-week course:
    • 10mg injected once a week for 4 weeks.
    • Followed by a maintenance dose of 5mg monthly (optional).
  • Injection: SubQ in the belly or thigh. It often leaves a red, itchy welt (histamine reaction) or bruise.

Primary Effects

1. Insulin Sensitivity

Drastically improves how the body handles carbohydrates.

2. Endurance

Increases the ability to perform physical work by optimizing cellular energy production.

3. Weight Control

Prevents fat gain even in the presence of a poor diet (in animal models).

Side Effects & Harm Reduction

Injection Pain:

PIP: MOTS-c is notorious for injection site pain. It burns going in and often leaves a welt that lasts for days.

1. Histamine Reaction

Localized redness and itching are very common. Taking an antihistamine 30 mins prior can help.

Tracking MOTS-c

Using the Anabolic Tracker app:

  1. Inventory: Add MOTS-c.
  2. Scheduling: Once Weekly.
  3. Metrics: Track Weight and Endurance (e.g., run times).
